    Transcranial Magnetic Stimulation

    More than 30 years have already passed since the seminal article of Baker et al., in which a transcranial magnetic stimulation (TMS) pulse over the motor cortex (MC) was successfully used to painlessly elicit a recordable electromyography response. Since then, multiple studies have been conducted exploring the underlying physiology of TMS effects, its potential uses in clinical settings, as well as safety. In this chapter, we provide an overview regarding the basic mechanisms of TMS, how it can be used as a tool to investigate human brain cortex physiology, how it can be used to alter cortical excitability, its safety concerns, and its use in clinical settings. © 2018 Elsevier Ltd.     Isolation of microglia and immune infiltrates from mouse and primate central nervous system

    Microglia are the innate immune cells of the central nervous system (CNS) and play an important role in the maintenance of tissue homeostasis, providing neural support and neuroprotection. Microglia constantly survey their environment and quickly respond to homeostatic perturbations. Microglia are increasingly implicated in neuropathological and neurodegenerative conditions, such as Alzheimer’s disease, Parkinson’s disease, and glioma progression. Here, we describe a detailed isolation protocol for microglia and immune infiltrates, optimized for large amounts of post mortem tissue from human and rhesus macaque, as well as smaller tissue amounts from mouse brain and spinal cord, that yield a highly purified microglia population (up to 98 % purity). This acute isolation protocol is based on mechanical dissociation and a two-step density gradient purification, followed by fluorescence-activated cell sorting (FACS) to obtain pure microglia and immune infiltrate populations. © Springer Science+Business Media LLC 2017.

    Reactive oxygen species and redox signaling in breast cancer

    Breast cancer progression depends on host innate and adaptive immune responses. Despite the effect of immune system in tumor destruction, cancer cells may induce the immune cells to support tumor progression. During tumor-host interactions, both tumor and immune cells produce significant amounts of reactive species (RS). The imbalance resulting from RS production and its neutralization is called oxidative stress, and it modulates several cellular processes impacting breast cancer outcome. Two major sensors of these processes are the nuclear factor erythroid-derived 2-related factor 2 (NRF2) and NF-κB-related pathways. The transcriptional factor NF-?B has prooxidant capacity, while Nrf2 has neutralizing action. NF-κB is a regulator of innate immunity, and it induces RS production and inflammatory cytokines. Furthermore, high level of NF-κB is found for breast cancer, and its downstream signaling has been implicated in aggressive tumor features. The redox sensor NRF2 is activated under oxidative stress conditions and induces human antioxidant response element in order to control cellular homeostasis; however, it protects tumors against RS caused by chemotherapy leading to treatment resistance. This chapter raises the discussion regarding the redox mechanisms involved in breast cancer development driven by the activation of the Nrf2-NF-κB axis. © 2016 by Taylor & Francis Group, LLC.

    Comparative metagenomics

    Thanks in large part to newer, better, and cheaper DNA sequencing technologies, an enormous number of metagenomic sequence datasets have been and continue to be generated, covering a huge variety of environmental niches, including several different human body sites. Comparing these metagenomes and identifying their commonalities and differences is a challenging task, due not only to the large amounts of data, but also because there are several methodological considerations that need to be taken into account to ensure an appropriate and sound comparison between datasets. In this chapter, we describe current techniques aimed at comparing metagenomes generated by 16S ribosomal RNA and shotgun DNA sequencing, emphasizing methodological issues that arise in these comparative studies. We provide a detailed case study to illustrate some of these techniques using data from the Human Microbiome Project comparing the microbial communities from ten buccal mucosa samples with ten tongue dorsum samples in terms of alpha diversity, beta diversity, and their taxonomic and functional profiles. © 2018, Springer Science+Business Media LLC.

    Caffeine and sport

    Caffeine is a trimethylxanthine found in coffee and several other foods and beverages. Its stimulatory effects make it an interesting strategy to boost performance for athletic populations. Scientific evidence supports its efficacy to improve high-intensity endurance exercise, explosive and high-intensity efforts, resistance exercise, team sports and combat sports, though individual variation in the ergogenic response to caffeine exists. Supplementation can be taken in many forms including dissolved in water, via capsules, coffee, energy drinks and caffeinated gum; ingestion via capsules, dissolved in water or in caffeinated gum appear to be most effective. Variability in the exercise response following caffeine supplementation may be explained by genetic factors or habitual caffeine consumption. Caffeine is an excellent supplement for athletes looking to improve their exercise performance, though some consideration of side-effects and impact on sleep are warranted. © 2023
